Abstract

Different numerical and structural chromosomal aberrations have been observed in cattle. Some of these chromosomal anomalies, such as XX/XY chimerism; X trisomy; mosaicism and Robertsonian translocation are associated with reproductive problems. The present study aimed to characterize the incidence of chromosomal alterations in cows used as embryo donors. Karyotypes of 63 Bos indicus and Bos taurus cows of different age and parity, maintained in uniform management conditions, were analyzed. The karyotype study was performed by means of metaphase analysis obtained after lymphocyte culture. Approximately 25 cells from each animal were analyzed and the diploid number 2n=60 was found in 62 animals. The karyotype analysis by conventional staining in 652 cells of the remaining animal showed the existence of 59,X0/60,XX/61,XXX mosaicism with the following percentages: X0 (1.23%), XX (97.85%) and XXX (0.92%). In a second culture using R-banding by 5-bromodeoxyuridine (BrDU), the same cell lineages were found in a 1:124:1 ratio, respectively. The animal carrying this karyotype presented a regular estrus cycles, having started 5 pregnancies, producing 3 viable calves, 1 still-born and, on the last pregnancy, a mummyfied fetus of about 6 months. The present information points out that, in the bovine, the chromosomal complement X0/XX/XXX does not occur exclusively in sterile females.
